Cities in the South 
We are travelling from Washington DC to Memphis at the end of September (about 2 weeks) and are going to hire an RV so that we can camp through the national parks in Virginia/North Carolina. Firstly, I can't find a rental company that will let me drop the RV off in Memphis or Nashville, so I think we'll have to drop it off in Atlanta instead and hire a car for the last few days. But anyway, what I really want advice on is that although we plan to camp in national parks where possible, I'd still like to at least see Richmond and Charleston (and maybe Savannah). Will travelling in an RV make this too difficult ?

That would depend on the size & type of RV, and what & where you want to see in those cities, getting around downtown, in an RV can be frustrating and difficult, and not all that enjoyable. Just a few things to consider are, traffic, street width,(do they park on both sides) places large enough to park an RV, overpass height, it would probably be better to park outside of the city & take public transportation, bus, taxi, train, subway, into town.
We did that in Boston, we stayed out near Bedford & took the subway into Boston.
